Chemical preparation
- Liste mit Substrates, Inhibitors, Uncouplers, ect.: MiPNet09.12_O2k-Titrations
Added sV Instructions to following pages
Same concentration for standard and small volume chamber
- Malate
- Glutamate
- Succinate
- Octanoylcarnitine
- ADP
- ATP
- Glucose: check Luis for the second paragraph that is not in titration list
- Glycerophosphate
Different concentration for different chambers
Concentrations prepared separately
- Pyruvate
- Ascorbate
- TMPD
- Cytochrome_c
- KCN ask Luiz or Marco for feasability (how much stock solution is needed, volume of aliquots...)
Conc for sV-chamber by dilution
- CCCP
- Rotenone
- Malonate
- Antimycin A
- Myxothiazol
- Sodium azide
- Carboxyatractyloside deleted higher concentration because not recommended any more
ToDo
- Oligomycin who works with Omy? There are two concentrations in the wiki, only the lower conc is on the titrations-pdf. Write instructions for both concentrations (and both chambers)? The second concentration is achieved by dilution - which dilution would be appropriate for the sV-concentration? Luiz: give instructions for the low conc only. The high conc is not recommended any more
- Salicylhydroxamic acid why is so much volume added for the standard chamber? is SHAM not soluble / too toxic to be more concentrated? And why is a much higher volume indicated for the sV-chamber titrations?
- Digitonin
- Catalase in MiR06
- Hydrogen peroxide
Questions
- I always indicated to take only part of the aliquot to dilute it, because in our aliquots you cannot rely on the volume - the last aliquot is usually not full. It would be more elegant if aliquots would have a defined volume or is it not feasible?
