Adenosine 5βdiphosphate (D), C10H15N5O10P2K, potassium salt; substrate of ANT and ATP synthase.
HRR: D: ADP**(Sigma,A 5285: Adenosine5βdiphosphate, C10H15N5O10P2K, potassium salt, contains 1 mol/mol H2O, MW=491.2 Preparation: 0.491 g/2 ml of final volume. Add 1 ml H2O and 450 ul 5 N KOH. ADP will dissolve. Check check pH and adjust to 7, adjust total volume to 2 ml and divide into 0.2 ml portions. Store frozen at -80 Β°C.
- To keep [Mg2+] constant during respiration measurement mix ADP with MgCl2 (0.6 mol/mol ADP).
Preparation of Mg-ADP: After adjusting pH of ADP to 7, add ? mg of MgCl2. White precipitate will occurr, which will be dissolved after 1-2 min of stirring on magnetic stirrer. Adjust final volume to 2 ml, divide into 0.2 ml portions and store at -80 Β°C.
