MitoPedia alert

MitoPedia
Did you know that if ET capacity (E) is lower than OXPHOS capacity (P, P>E) in living cells or mitochondrial preparations, then you have encountered an experimental artefact? The possible triggers of this experimental artefact are:

- loss of oxidative capacity during the time course of the respirometric assay,
- insufficient uncoupler concentrations,
- over titration of an uncoupler, which inhibits ET capacity,
- inhibition of ET capacity due to the use of high oligomycin (inhibitor of the ATP synthase) concentrations employed to measure LEAK respiration prior ET capacity.

Antunes Diana (talk) 11:13, 22 June 2020 (CEST) Comments and suggestions relevant for checking:

  • For 'insufficient uncoupler concentrations' would not be more accurate 'insufficient uncoupler titrations' since we recommend always a titration approach with small volumes to avoid over titration? Maybe by writing 'insufficient uncoupler concentrations' it would lead the user towards using high U concentration per titration instead of using low volumes in each titration?
  • include relevant links (e.g., for ce and mtprep)?
  • Not sure if you agree in including the 'E' and 'P' icons within the alert - I thought it would be good just for a quick visualization of the terms.

Suggestion 2:

  • While in Home Office: Gnaigere (talk) 14:03, 19 March 2020 (CET) https://wiki.oroboros.at/index.php/Talk:O2k-Open_Support_alert - Many labs will move to home office mode. In line with our special DL7.4 home office offer (see Agenda today), therefore, I suggest to post in our Agenda analysis-linked O2k-Open Support cases during this time, and collect real-time operation cases for future releases.

O2k-Open SupportWarning: loss of data Find out how to evaluate the effect of the obtained ‘O2 slope neg’ noise on experimental results

- » QC1: Oxygen sensor test«

or

O2k-Open SupportWarning: loss of data How to decide whether the noise of the O2 slope neg. is within recommended values?

- » Noise of 'O2 slope neg.'«

(NOTE for 4-eyes: QC1: Oxygen sensor test is the O2k-Support page for sensor test, and Noise of 'O2 slope neg.' is the updated version by Erich that belongs to MitoPedia:_DatLab#Calibration. What to do with this pages? Shall the info remain in those locations or shall be linked instead?)

O2k-Open SupportWarning: loss of data Do you perform the leaky chamber test after chamber assembly?

An unnoticed leak from the chamber may affect your data, therefore we recommend performing the Leaky chamber test - especially after chamber reassembly.

New O2k-Open SupportWarning: loss of data A damaged OroboPOS-Holder may cause medium to leak from the O2k-Chamber.

See how you can check that the two blue OroboPOS-Holders are intact.

New O2k-Open SupportTake care Calibration parameters are transferred to experimental DatLab files

Save calibration information from selected Marks while DatLab is connected to the O2k during an instrumental calibration run. Air calibration and zero oxygen calibration can be performed and saved in a single DatLab file or in separate DatLab files.
- » Oxygen calibration - DatLab«

O2k-Open SupportWarning: loss of data Do you perform O2k-Chamber volume calibration?

The volume calibration is essential to ensure that the chamber volume is correctly calibrated to its standard volume of 2 mL (or 0.5 mL for the O2k-sV-Module).
- »O2k-Chamber MitoPedia«

O2k-Open SupportTake care Do you use the correct O2 solubility factor (FM) in the ‘O2 calibration’ window in DatLab?

The O2 solubility factor varies according to the medium used and influences the air calibration.
