Abstract: The two main goals of this thesis were to set up a method for measuring the oxygen consumption of isolated brown fat cells and to assess the difference in oxygen consumption of brown adipocytes between alpha-2-adrenoceptor-A, –AC knock-out and wild-type mice. In earlier studies a difference in weight gain between α2A-KO, α2AC-KO and wild-type mice was observed and the hypothesis was that the difference might be due to different metabolic activities of the brown fat. The project was carried out at Turku University Institute of Biomedicine, Department of Pharmacology, during summer 2012. An Oxygraph-2K high resolution respirometer was used to measure the oxygen consumption in isolated mouse brown fat cells obtained from the suprascapular brown fat depots. The Oxygen consumption of the isolated cells was measured at the basal level and after noradrenaline stimulation. Test groups were formed of a total of 32 mice both male and female, 5-6 mice in each group. All mice had been fed a high fat diet to make them over weight. The results were analyzed using 2-way ANOVA. The results suggest that the brown fat metabolic activity of α2A-KO mice had altered. The baseline oxygen consumption was significantly lower and the noradrenaline response was higher than with the other mice. Whether this is due to the larger weight gain or vice versa cannot be said based on this study. Additional studies should be conducted with lean normal weight mice or alternatively by stimulating the noradrenaline production of the overweight mice in vivo with beta-receptor agonists. The setting up of the Oxygraph-2K and the method for measuring the oxygen consumption of isolated brown fat cells was successful and might be used for similar tests later on.
