A rapidly increasing number of human pathologies is linked to mitochondrial dysfunction. During the past few years, blood cells such as peripheral blood mononuclear cells (PBMC) and platelets have become the targets in investigations on various pathologies. However, standard operating procedures for mitochondrial respiratory studies on blood cells are still missing. Application of different anticoagulants, different isolation methods and storage, and different respiratory protocols make it impossible to compare results on a quantitative basis. In our preliminary literature analysis, >50 publications on mitochondrial respiration of PBMCs were compared in terms of methodology of cell isolation and mitochondrial respiration. More than eight cell isolation protocols were used, but incomplete description of the methods renders replication of the experiments largely impossible. More than 10 different mitochondrial respiration media were used. As a result, the data on mitochondrial respiration showed a very wide scatter. Harmonization of protocols by direct determination of the effect of various procedures on the results will provide a basis to ‘translate’ different studies and improve their comparability. Application of standardized methods for isolation and conditions for respirometric measurements would give basis for comparability of results in translational research.
