Abstract: The objective of this study was to evaluate the radical scavenging activity and the antioxidant effect of Allium sativum bulb variety Huaralino by chemical and 8 biochemical in vitro techniques.
The bulb extract was prepared in double distilled water at concentrations of 20 - 40 mg/mL. Assays for studying sulfur secondary metabolites and their properties were quantification of allicin and organic compounds with thiol group; and the formation of nitrosothiols. For oxygenated metabolites were evaluated polyphenols and flavonoids. The ability to capture free radicals was determined by the techniques of ABTS.+ and DPPH radicals also capture of superoxide, hydrogen peroxide decomposition and FRAP; all of them for spectrophotometry. Mineral was also quantified by atomic absorption. Assays for in vitro antioxidant activity study made by carbonyl formation in albumin treated with hydrogen peroxide and TBARS levels in red blood cells also treated with hydrogen peroxide. Determination of V3, V4 and ICR was performed on isolated mitochondria of rat liver by polarography.
The main results of sulfur and oxygenated metabolites were: a) allicin 2,8 mg/g of fresh peeled bulb, b) low molecular weight thiols μmol 0.95 Cys/g dry mass, c) 2.52 mg polyphenols EAG/g dry mass d) EQ 1.71 mg/g dry mass. The free radical scavenging was made by hydrogen donation, donating and electron withdrawing, superoxide uptake, decomposition of hydrogen peroxide and ferric iron reduction. The extract has antioxidant protection decreasing significantly (p<0.05) the formation of carbonyl and TBARS levels. ICR also decreases depending on the concentration of the extract.
In conclusion, the aqueous extract of Allium sativum bulb variety Huaralino has sulfur and oxygenated antioxidant metabolites capable of capturing free radicals which exert antioxidant effect in vitro, comparable with the international literature. • Keywords: Allium sativum, Free radicals, Antioxidant activity, Secondary metabolites
